摘 要:利用海枣曲霉发酵获得菊糖酶粗酶液,对粗酶液的发酵条件和酶解条件进行了研究。结果表明,菊糖酶的最适发酵条件为:菊糖2%,磷酸二氢铵0.5%,磷酸二氢钾0.25%,MgSO4.7H2O 0.05%,pH 6.0-7.0,接种量8%,温度28℃,时间60 h,按最优条件得到的菊糖酶粗酶液,酶活为18.39 U/mL,I/S值为45.71。菊糖酶的最适酶解条件:温度60℃,加酶量11.0 U/g菊糖,水解12 h,底物浓度10%,pH 6.0-7.0。菊糖酶解率最高达59.3%。运用超滤法纯化菊糖酶解产物得到低聚果糖,利用高效凝胶渗透色谱法(HPGPC)测定其相对分子质量为509,聚合度为2-3。Inulinase was produced by Aspergillus phoenicis-mediated fermentation.The conditions of fermentation and enzymolysis were studied.The results showed that the optimal fermentation conditions were: inulin 2%,NH4H2PO4 0.5%,KH2PO4 0.25%,MgSO4.7H2O 0.05%,pH 6.0-7.0,inoculum size 8%,temperature 28 ℃ and fermentation time 60 h.Under these conditions,the enzyme activity and I/S of inulase were 18.39 U/mL and 45.71,respectively.The optimal enzymolysis conditions were: temperature 60 ℃,additive amount of enzyme 11.0 U/g inulin,enzymolysis time 12 h,concertration of substrate 10%,pH 6.0~7.0.The rate of inulin enzymolysis was 59.3% under the optimal conditions.High-purity fructooligosaccharide was obtained by ultrafilter device and its molecular weight and polymerization degree were determined by HPGPC as 509 and 2-3,respectively.
