摘  要:以真鳕鱼(Gadus macrocephalus)鱼皮为原料,采用酸、碱、盐和酸碱联合处理4种不同预处理方式,经酶解、过滤、脱色脱腥、冻干等工序制备胶原蛋白肽,比较不同预处理制得胶原蛋白肽的得率和灰分、盐分、金属元素含量以及氨基酸组成。结果表明,不同预处理制得的胶原蛋白肽灰分、盐分含量均为碱处理<盐处理<酸处理<酸碱联合处理,差异均极显著(p<0.01);金属元素中主要是钠离子和钙离子,占12种金属元素的95.4%以上。0.1%NaOH预处理制备的胶原蛋白肽得率、氨基酸总量及羟脯氨酸占总氨基酸的比例最高,分别为15.48%,91.87%,10.48%。因此,用碱预处理真鳕鱼皮制备的胶原蛋白肽纯度较高。Codfish (Gadus macrocephalus) skin is used as the raw materials in this study. Using different pretreatment methods of acid, alkali, salt, and combined treatment with acid and alkali, the collagen is isolated from codfish skin by the processes of enzymatic hydrolysis, filtration, decoloration deodorization, freeze dry. The yield, ash, salt, metal elements and amino acids of four group collagen peptides are compared and analyzed. Results show that the order of ash and salt in collagen peptides prepared from codfish skin by different pretreatment methods is alkali treatment&lt;salt treatment&lt;acid treatment&lt;combined treatment with acid and alkali, the difference is extremely significant (p&lt;0.01) . Sodium and calcium ions accounted for more than 95.4% of the total of 12 kinds of metal elements, which are the main metal element in four collagen peptides. Collagen peptide extracted from codfish skin by 0.1% NaOH pretreatment has the highest yield, total amino acids content, and also has the highest proportion of hydroxyproline in total amino acids, respectively for 15.48%, 91.87%, 10.48%. Therefore, collagen peptide extracted from codfish skin by alkali pretreatment has higher purity.
