摘 要:本文采用响应面法对利用木糖产普鲁兰糖发酵培养基进行了优化。Plackett-Burman实验筛选出影响普鲁兰糖产量的三个主要因素,分别为木糖、硫酸铵和磷酸氢二钾。在此基础上,利用中心复合实验和响应面分析确定了主要影响因素的最佳浓度,即木糖55g/L、硫酸铵0.7g/L、磷酸氢二钾5.8g/L。摇瓶验证实验表明,优化后普鲁兰糖产量达到16.25g/L,较优化前提高了27.55%。傅里叶红外光谱分析发现,获得的普鲁兰糖同普鲁兰糖标品结构基本相同。本研究有效提升了普鲁兰糖产量,为规模化利用纤维素类物质生产普鲁兰糖奠定了基础。In this study, pullulan fermentation medium was optimized through response surface methodology.Thethree main factors that played important roles in pullulan production which identified by Plackett-Burman design,were xylose, (NH)2SO4 and K2HPO4. Subsequently, the central composite design and response surfacemethodology were applied to further investigate the optimum concentration of the main factors. The resultsindicated that the optimized concentrations of the main factors were.xylose 55g/L, (NH)2SO4 0.7g/L and K2 HPQ5.8g/L.With the optimized medium, maximal pullulan obtained in the shake flasks was 16.25g/L, of which increased27.55% compared with the fermentation before optimization.IR spectra of the samples indicated that,the structureof the purified pullulan and the pullulan standard was identicaI.Pullulan production was increased significantly inthis study.It laid a basis for the scale up of pullulan production from cellulosic materials.
