摘  要:以山葡萄酿酒废弃物为原料,酶法提取可溶性膳食纤维。采用Box-Behnken设计方法对纤维素酶用量、半纤维素酶用量、提取时间和提取温度4个单因素工艺参数进行优化,用响应面分析法确定4个因素的最佳工艺条件,建立数学模型。结果表明:纤维素酶用量为93 IU/g、半纤维素酶用量为121 IU/g、提取时间为257 min和提取温度为51℃,在此条件下进行验证试验,可溶性膳食纤维提取率为152.13 mg/g。山葡萄渣可溶性膳食纤维功能特性的试验结果表明,对DPPH自由基的清除率超过50.31%,膨胀力和持水力分别为5.57 mL/g和4.91 g/g。Soluble dietary fiber was prepared from wild grape pomace using enzymatic method. Based on Box-Behnken design, cellulase concentration, hemicellulase concentration, extraction time and extraction temperature were chosen as the four important factors with three levels. The process conditions were optimized by response surface method, and a mathematical model was setted up. The optimized parameters for soluble dietary fiber extraction were as follows: cellulase concentration 93 IU/g, hemicellulase concentration 121 IU/g, extraction time 257 min and extraction temperature 51 ℃. The yield rate of soluble dietary fiber reached 152.13 mg/g. The function properties of soluble dietary fiber from wild grape were investigated. Scavenging rate of DPPH radicals exceeded 50.31%. The swelling capacity was 5.57 mL/g and water holding capacity was 4.91 g/g.
