摘  要:利用废弃的青鱼鱼鳞制备胶原蛋白肽,不仅能够减少环境污染,还可以明显提高鱼类加工废弃物的综合利用,提高经济效益。采用青鱼鱼鳞为原料,通过测定胶原蛋白溶出率及脱钙率,优化温水清洗及盐酸脱钙的预处理条件;测定酶解液水解度及低聚肽占成品百分含量,确定并优化复合酶水解的条件,得到所需的肽。最佳工艺条件:(1)采用料液比为1∶15的50℃温水水洗2 h,可得表面无杂质、胶原蛋白溶出率仅为2.2%的鱼鳞;(2)在0.4 mol/L盐酸与料液比为1∶10条件脱钙30 min,脱钙率达94%,而其工艺过程中的胶原蛋白溶出率仅有0.15%;(3)采用2 000 U/g的木瓜+菠萝+碱性蛋白酶(1∶1∶2),50℃自然p H下酶解5 h,获得水解度47%的胶原蛋白低聚肽,其分子质量主要分布在180~1 000 u和1 000~2 000 u两段,含量分别为73.3%和16.6%。成品胶原蛋白低聚肽颜色为白色泛微黄,具有很好的溶解性。Recently, it is of interest to develop collagen peptides derivative from scales(e.g: herring fish), which is a type of byproducts, has received considerable attention. The processing reduces the environmental pollution, and remarkably contributes enhanced value in fish industry. Herring fish scales chosen as raw materials, we optimized pretreatment conditions by determining decalcification and collagen dissolution rate, regulated the combined enzymatic hydrolysis conditions via determining degree of hydrolysis and yield of oligopeptide, and normally harvested desirable peptides.Overall, the optimal processing condition for preparation of scales peptides was the sequential combination of as follows. (1) washing: 1:15, the ratio of feed liquid, conducted at 50℃ for 30min, then subjected to surface clean, and obtained 2.2% collagen dissolution rate; (2) demineralization: 0.4 N HC1 applied with the rate of 1:10 for 30 min soaping, received 94% decalcification rate; (3) enzymatic hydrolysis: the ratio was 1:1:2, both at the E/S 2 000 U/g, hydrolyzed at 50 ℃ and pH 6 for 5 h, acquired 47% degree of hydrolysis of collagen-based peptide, which covers two main fractions at 180-1 000 u and 1 000-2 000 u with ratio 73.3% and 16.6%, respectively. The final products were of pale but yellowish collagen peptides with merit of the property of high solubility. In a conclusion, this kind of novel technology provides a potent way to produce scales-based peptides with merits of potential application in practical food production.
