摘 要:目的:体外和动物实验研究表明,玉米肽(CPs)可通过增加乙醇脱氢酶(ADH)和乙醛脱氢酶(ALDH)的活性促进乙醇代谢。但酶活性的增加是通过改变催化效率还是改变酶的生成量迄今仍不清楚。本研究的目的是确定CPs能否改变酶蛋白的数量从而调节人体酒精代谢。方法:60名健康男性青年志愿者被要求连续4 d每天摄入含有30g乙醇的白酒,玉米肽组(n=30)在第2、3、4日饮酒之前分别口服1g、2g、4g CPs。乙醇对照组(n=30)4 d中饮酒前都不给予玉米肽。喝完最后一口酒30min后抽取静脉血,测定全血乙醇(BAC)、血清乙醇脱氢酶(ADH)、血清乙醛脱氢酶(ALDH)的含量,并检测饮酒后3 h内呼气乙醇水平(BrAC)的变化。结果:与第1日(乙醇对照)比,酒后30 min的BAC在口服2g、4g CPs的这两日,分别显著降低9.5%、23.7%。同样,口服2g、4g CPs也显著降低了酒后30 min的BrAC(P〈0.05和0.01)。此外,口服4g CPs这一日所有时间点(30-180 min)的BrAC均显著降低。然而,不同剂量CPs的摄入都没有显著改变血清ADH和ALDH的水平。结论:玉米肽降低酒后BAC和BrAC的水平存在一定的剂量-效应关系,提示CPs能够促进健康青年男性的乙醇代谢。但CPs对乙醇代谢影响的潜在机理需要进一步研究。Objective In vitro and animal studies have suggested that corn peptides (CPs) facilitate alcohol metabolism by increasing the activities of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH). It remains unknown whether the activities of these enzymes were increased by altering the catalytic efficiency or the amount of enzyme produced. The objective of the present study was to determine if CPs can alter the amount of enzyme proteins and modulate alcohol metabolism in humans. Methods A total of sixty healthy young Chinese men were recruited and asked to consume 30 g of alcohol on four consecutive days. 1, 2 and 4 g of CPs were administered orally immediately before alcohol consumption in CPs group (n=30) on days 2, 3 and 4, respectively. NO CPs were given for alcohol control group (n=30) on all 4 days. Results CPs ingestion was demonstrated to lower both BAC and BrAC in a dose-dependent pattern after alcohol consumption in CPs group. The intake of 2 and 4 g of CPs significantly decreased the BAC at 30 min post-consumption by 9.5% and 23.7%, respectively, compared with the alcohol control. In accordance, the intake of 2 and 4 g of CPs was also shown to significantly decrease BrACs at 30 min (P〈0.05 and 0.01, respectively). Furthermore, significantly lowered BrACs persisted at all time points (30-180 min) in the day that received 4 g of CPs. However, no significant changes in serum levels of ADH and ALDH were observed in any of the days in CPs group. Conclusion The study confirmed that CPs can facilitate alcohol metabolism in young men, but this effect was not mediated by increasing the amount of ADH and ALDH. The mechanisms underlying the effects of CPs on alcohol metabolism require further study.
