摘  要:采用Alcalase蛋白酶酶解从玉米脱脂蛋白粉中分离得到的玉米谷蛋白,对酶解产物采用超滤、凝胶过滤层析和高效液相色谱法分离纯化玉米谷蛋白抗氧化肽,并研究其抗氧化活性。结果表明,Alcalase蛋白酶酶解玉米谷蛋白获得的抗氧化肽分子量主要集中在低于1 kDa,其DPPH自由基和羟自由基清除率分别为80.46%、81.36%。凝胶过滤层析得到抗氧化活性较高的组分P6、P7和P8,其DPPH自由基清除率分别为32.05%、24.43%、14.19%,多肽含量分别为0.638、0.253、0.158 mg/mL。高效液相色谱对这3个组分进一步分离纯化分别得到主要组分。The corn glutelin extracted from corn gluten meal was hydrolyzed by alcalase protease. The hydrolysates were separated and purified by ultrafiltration, gel filtration chromatography and high performance liquid chromatography(HPLC) to obtain antioxidant peptides, and their antioxidant activities were studied. The results indicated that the small-sized peptides (Mw 〈 1 kDa) which were the major fractions of the hydrolysates possessed higher antioxidant activities, and their DPPH radical scavenging rate and hydroxyl radical scavenging rate were 80.46%and 81.36%, respectively. P6, P7 and P8 from gel filtration chromatography possessed higher antioxidant activities with DPPH radical scavenging rate of 32.05%, 24.43% and 14.19%, respectively, and their peptides contents were 0.638, 0.253 and 0.158 mg/mL, respectively. Finally, their major fractions were obtained from P6, P7 and P8 by reserved-phase high performance liquid chromatography (HPLC).
