摘 要:皮蛋是我国独创的一种传统大宗蛋制品,也是一种良好的“清热去火”食品。以“去大肠火”为导向,运用TNF-α诱导的Caco-2细胞炎症反应模型和DSS诱导的小鼠结肠炎模型考察皮蛋蛋白水提物的体内、外抗肠炎活性。结果表明:皮蛋蛋白水提物(5和10mg/mL)显著抑制Caco-2细胞IL-8的分泌,下调促炎细胞因子IL-8、TNF-α、IL-6和IL-1β基因的表达,并上调抗炎因子IL-10基因的表达。皮蛋蛋白水提物(150或300mg/kgBW)可以有效缓解DSS诱导的结肠炎小鼠体重减轻和炎症临床症状,降低促炎细胞因子TNF-α和IL-6的分泌,并下调结肠中促炎细胞因子TNF-α、IL-6、IL-1β、IFN-γ、MCP-1和IL-17A基因的表达。体内、外试验结果说明皮蛋蛋白水提物具有抗肠炎活性,可有效保护肠道健康。Preserved egg is one of the original traditional bulk egg products, and is also a kind of good “clear heat” food. Oriented to anti-inflammatory effects on intestinal system, the effects of water extract of preserved egg white on Caco-2 cells induced by TNF-α and a mouse model of dextran sodium sulfate (DSS)-induced colitis were evaluated. The results showed that water extract of preserved egg white inhibited the secretion of IL-8, decreased the expression of the pro-inflammatory cytokines TNF-α, IL-6, IL-1β, IFN-γ and IL-17, and increased the expression of IL-10 from Caco-2 cells. The treatment of extract of preserved egg white (150 or 300 mg/kg BW) reduced DSS-induced colitis symptoms, improved colon histology, reduced secretion of pro-inflammatory cytokines TNF-α and IL-6, and decreased gene expression of the pro-inflammatory cytokines TNF-α, IL-6, IL-1β, IFN-γ and IL-17 in the colon. The experimental results in vivo and in vitro showed that water extract of preserved egg white exerts anti-inflammatory effects on intestinal system, which can effectively protect the intestinal health.
