摘  要:【目的】研究高压脉冲电场对普洱熟茶中茶多糖和茶多酚抗氧化活性的影响,为提高普洱熟茶体外抗氧化活性提供一种有前景的原料处理方法。【方法】利用不同条件的高压脉冲电场(high voltage pulsed electric field, HPEF)对普洱熟茶茶样进行处理,提取茶样茶多糖和茶多酚,以ABTS自由基清除能力为指标,测定茶多糖和茶多酚的抗氧化活性。利用Matlab对茶多糖与茶多酚的抗氧化活性进行逐步回归分析,建立其与HPEF电压及频率之间的数学模型,并绘制模型三维关系图,研究分析不同条件HPEF对茶多糖与茶多酚抗氧化活性的影响。【结果】影响茶多糖对ABTS自由基的清除率最主要的HPEF参数是电压,且当电压为12 kV、频率为120 Hz左右时,茶多糖对ABTS自由基的清除作用最强。影响茶多酚对ABTS清除能力最主要的HPEF参数是频率,频率为140 Hz左右时,茶多酚对ABTS自由基的清除作用效果最强。【结论】合适条件的HPEF处理能在一定程度上提高普洱熟茶茶多糖和茶多酚的抗氧化活性,本研究对普洱茶天然抗氧化剂的开发有一定的借鉴意义。[Purpose]The effects of high voltage pulsed electric field on the antioxidant activity of tea polysaccharides and polyphenols in Pu'er ripe tea were studied in order to provide a promising raw material treatment method for improving the antioxidant activity of Pu'er ripe tea in vitro.[Method]We used different high voltage pulsed electric fields (HPEF) to deal with the third grade Pu'er ripe tea, extract the tea polysaccharides and tea polyphenols and determine antioxidant activity of tea polysaccharides and tea polyphenols in each tea sample by ABTS radical scavenging rate. Then antioxidant activity of tea polysaccharides and tea polyphenols were analyzed by stepwise regression analysis on Matlab, we established the mathematical model of its antioxidant activity and voltage, frequency, and drew the three-dimensional relationship diagram of the model to study the effect of different HPEF on the antioxidant activity.[Results]The most important HPEF parameter affecting the scavenging ability of tea polysaccharides on ABTS was voltage, and when the HPEF voltage was 12 kV and the frequency was about 120 Hz, the scavenging effect of tea polysaccharide on ABTS radical was the strongest. The most important HPEF parameter affecting the scavenging ability of tea polyphenols on ABTS was frequency, and when the frequency was about 140 Hz, the scavenging effect of tea polyphenols on ABTS radical was the strongest.[Conclusion]The suitable conditions of HPEF can improve the antioxidant activity of tea polysaccharides and tea polyphenols in Pu'er tea to a certain extent. This study provides a reference for the development of natural antioxidants in Pu'er tea.
