三种红曲菌固态发酵产桔霉素及桔霉素生物合成相关基因的比较

摘  要:采用高效液相色谱-荧光检测法测定了3种22株红曲菌所产红曲中的桔霉素含量,并用2对引物通过PCR和测序的方法检测了这22株红曲菌的桔霉素生物合成相关基因pksCT和ctnA的分布情况,探讨了两者的相关性,旨在为筛选低产甚至不产桔霉素的红曲菌菌株提供信息。研究表明,11株紫色红曲菌和4株红色红曲菌可产生桔霉素,pksCT和ctnA扩增阳性;6株红色红曲菌未检出桔霉素,pksCT扩增阳性,但ctnA扩增阴性;1株血红红曲菌的pksCT在扩增的编码区有1个单碱基缺失,且ctnA扩增阴性,未检出桔霉素。结果表明,基因组中同时存在有功能的pksCT和ctnA基因与3种红曲菌的固态发酵产物桔霉素检测阳性结果相关。In this study,the citrinin content of fermented rice produced by 22 strains from three species of Monascus were analyzed using high performance liquid chromatography with fluorescence detection,and two pairs of primers were used to amplify the citrinin biosynthesis related genes of these strains by polymerase chain reaction.The correlation between citrinin production and the key genes responsible for citrinin biosynthesis in these strains was investigated.All 11 M.purpureus strains and four M.ruber strains could produce citrinin,and the amplification of both pksCT and ctnA was positive.The amplification of pksCT was positive,while the amplification of ctnA was negative in the other six M.ruber strains with the phenotype of undetectable citrinin production.A single-base deletion was observed in the amplified pksCT coding regions of M.sanguineus R18,and both ctnA amplification and citrinin detection were negative.It was indicated that the simultaneous presence of functional pksCT and ctnA genes was highly related to citrinin production among M.purpureus,M.ruber and M.sanguineus.
