摘  要:利用酶工程技术,以鱿鱼皮为原料,研究制备鱿鱼皮胶原蛋白肽粉。选用胃蛋白酶、中性蛋白酶、木瓜蛋白酶、胰蛋白酶和alcalase蛋白酶进行试验,以产品得率和肽及氨基酸含量为评价指标,从5种蛋白酶中筛选中性蛋白酶作为水解用酶。进行不同料液比、不同加酶量和不同酶解时间的单因素试验,初步确定料液比1︰6 (kg/L),加酶量600 U/g,酶解时间6 h。在此基础上进行加酶量、温度、pH和时间的四因素三水平正交试验,确定最佳酶解条件:加酶量600 U/g,温度45℃, pH 6.0,酶解时间4 h。酶解得到的胶原蛋白肽粉呈淡黄色粉末状,糖含量为3.9%,总脂肪含量为0.19%,蛋白含量为73.57%,肽粉中的羟脯氨酸含量为6.35%,说明该肽粉营养价值较高,达到胶原蛋白肽粉的行业标准要求,在实际生产中有广阔的应用前景。The preparation of squid skin collogen peptide powder was studied using enzyme engineering technology. Pepsin,neutral protease, papain, trypsin and alcalase were selected for the experiment, and the neutral protease was chosen as the hydrolysis enzyme by evaluating the yield of the product and the content of peptide and amino acid. The single factor experiment of different solid-liquid ratio, enzyme dosage and enzymatic hydrolysis time was conucte. The results showed that the solid-liquid ratio was 1︰6(kg/L), the enzyme dosage was 600 U/g, and the enzymatic hydrolysis time was 6 h. On the basis of the above experiments, the orthogonal experiment of four factors about enzyme dosage, temperature, pH and time and three levels was carried out. The optimal enzymolysis condition was enzyme dosage 600 U/g, temperature 45 ℃, pH 6.0 and enzymolysis time 4 h. The collagen peptide powder was light yellow powder with 3.9% sugar, 0.19% total fat, 73.57% protein and 6.35%hydroxyproline. The peptide powder had reached the industrial standard requirements of collagen peptide powder, and had wide application in future.
