摘  要:为筛选抑菌性较强的植物精油并优化精油-β-环糊精微胶囊的制备工艺,采用滤纸片法,测定14种植物精油对假单胞菌、铜绿假单胞菌、大肠杆菌和金黄色葡萄球菌的敏感程度,以筛选出抑菌性最强的精油为芯材、β-环糊精为壁材,制备微胶囊。在单因素试验基础上,以包埋率为响应值,采用响应面法优化工艺条件。以抑菌率为指标,测定微胶囊的抑菌性能。结果表明,百里香精油对4种供试菌高度敏感。百里香精油-β-环糊精微胶囊最佳制备条件为壁芯比9.3︰1(g/g)、包合时间1.5 h、包合温度42℃,在此条件下制备的微胶囊包埋率为71.01%±1.21%,预测值与实测值相对误差为2.61%。精油净含量相同的干态与湿态微胶囊对金黄色葡萄球菌抑菌率最高,其次是大肠杆菌,最低的是假单胞菌和铜绿假单胞菌,且干态微胶囊抑菌率均显著低于湿态(P<0.01)。响应面法优化的百里香精油-β-环糊精微胶囊制备工艺可行,为植物精油的开发利用提供参考。In order to screen plant essential oil with strong antibacterial property and to optimize the preparation process of essential oil-β-cyclodextrin microcapsule,the sensitivity of 14 plant essential oils to Pseudomonas,Pseudomonas aeruginosa,Escherichia coli and Staphylococcus aureus was determined by filter paper method.The microcapsule was prepared by selecting the most antibacterial essential oils as core material andβ-cyclodextrin as wall material.On the base of the single factor experiment,response surface method was used to optimize the process conditions with the embedding rate as the response value.The bacteriostatic performance of microcapsule was determined by the bacteriostatic rate.The results showed that thyme essential oil was highly sensitive to four tested bacteria.The optimal preparation condition was as follows:wall-coreratio 9.3︰1(g/g),inclusion time 1.5 h,and inclusion temperature 42℃.The embedding rate of microcapsule prepared under this condition was 71.01%±1.21%.The relative error between the predicted value and the measured value was 2.61%.The dry and wet microcapsules with the same net content of essential oil had the highest antibacterial rate against Staphylococcus aureus,followed by Escherichia coli,and the lowest antibacterial rates were Pseudomonas and Pseudomonas aeruginosa.The antibacterial rate of dry microcapsule was significantly lower than that of wet microcapsule(P<0.01).The preparation process of thyme essential oil-β-cyclodextrin microcapsules optimized by response surface methodology is feasible,which provides a reference for the development and utilization of plant essential oil.
