摘 要:以短小芽孢杆菌1.894(Bacillus pumilus1.894)为出发菌株,经紫外诱变育种,得到了产碱性蛋白酶较高的新菌株2080;并对其液态发酵条件和部分酶学特性进行了研究。结果表明,其较优的培养基组成为:菜粕1.5%,干酪素1%,D-果糖1%,KH2PO40.1%,MgSO·47H2O0.02%,CaCl·22H2O0.01%,pH8.5;接种后,30℃,发酵84h,酶产率可达4.04×106U/L。该酶的最适pH为10,最适温度为50℃,分别在pH8~10和30℃~45℃范围内较稳定。A higher yield strain Bacillus pumilus 2080 was obtained by UV mutagenesis from Bacillus pumilus 1.894 for the production of alkaline protease. Subsequently, submerged fermentation (SmF) conditions and partial characterizations of alkaline protease from the mutant strain were investigated. The optimal medium contained(%): rapeseed meal 1.5, casein 1, D-fructose 1, KH2PO4 0.1, MgSO4·7H2O 0.02, CaC2l·2H2O 0.01, pH 8.5. After inoculated and incubated at 30 ℃ for 84 h by SmF, the maximum alkaline protease yield of 4.04×10^6 U/L was obtained from Bacillus pumilus 2080. The highest activity of the crude enzyme from Bacillus pumilus 2080 is at pH 10 and 50 ℃ respectively. The enzyme was stable not only from pH 8 to pH 10 at 40 ℃ for 30 min, but also from 30℃~45 ℃ for 30 min in Na2CO3/NaHCO3 buffer(0.1 mol/L, pH10).
