摘 要:用低聚壳聚糖、低聚半乳糖、低聚异麦芽糖、壳聚糖和羧甲基壳聚糖对牛乳β-乳球蛋白进行糖基化处理,通过动物实验及ELISA方法检测各糖基化产物的抗原性和免疫原性,结果显示低聚半乳糖的糖基化处理降低牛乳β-乳球蛋白的抗原性和免疫原性效果最佳,分别降低41%和18%。凝胶过滤层析和SDS-聚丙烯酰胺凝胶电泳分析结果显示,低聚半乳糖糖基化产物的相对分子质量为40kDa,保持原有的二聚体结构。该结合物与β-乳球蛋白比较,乳化稳定性、溶解性和分散性无显著性差异,而热稳定性和抗氧化性明显提高。以上结果表明,低聚半乳糖是糖基化法降低牛乳β-乳球蛋白免疫原性的较理想的糖基供体。Bovine milk β- lactoglobulin was conjugated with chitooligosaccharide, galactooligosaccharide, isomaltooligosaccharide, chitosan and carboxymethylchitosan, respectively. Antigenicity and immunogenicity of each product were measured by animal experiment and ELISA assay.The results showed that conjugation with galactooligosaccharide was the most effective method on reducing antigenicity and immunogenicity of β-Iactoglobulin (decreased for 41% and 18%, respectively).The results of gel filtration chromatography and SDS-PAGE assay indicated the molecular weight of conjugate of galactooligosaccharide was 40kDa,and was still maintained in dimmer form.Galactooligosaccharide conjugate was no distinct difference in emulsification stability, solubility and dispersion, whereas was improved in thermal stability and antioxidant ability compared with β-lactoglobulin. ln conclusion, galactooligosaccharide is a better glycosyl donor for reducing immunogenicity of bovine milk 6-LG by glycation.
