摘  要:目的:制备左旋和和右旋虾青素,明确不同立体构型虾青素抗脂质过氧化能力。方法:从不同原料中分离纯化虾青素,构建亚油酸、肝线粒体和红细胞氧化模型。通过检测不同指标来评价不同立体构型虾青素抗脂质过氧化活性。结果:从雨生红球藻和红法夫酵母中制备的虾青素分别为左旋和右旋。3种立体构型虾青素对亚油酸自动氧化程度和Fe2+诱导MDA生成、AAPH诱导线粒体MDA生成、线粒体肿胀和蛋白质羰基含量、H2O2诱导红细胞溶血率均有明显的抑制作用。左旋和右旋虾青素抗脂质过氧化能力显著高于混合型,高浓度情况下,左旋虾青素在线粒体MDA抑制率和线粒体肿胀抑制率方面显著高于右旋虾青素。3种立体异构体虾青素在氧化条件下的抗氧化酶(SOD和GSH-PX)活力没有显著差异,表明抗氧化酶系统不是造成不同立体构型虾青素抗脂质过氧化能力存在差异的原因。结论:不同立体构型虾青素抗脂质过氧化活性存在差异,为合理开发虾青素功能产品提供了理论依据。The objectives of this study were to prepare 3 S,3′S(S), 3 R,3′R(R) astanxanthin(AST) and determine the anti-lipid peroxidation activity of different stereoisomeric AST. AST were separated and purified from different raw materials. The anti-lipid peroxidation activity of different stereoisomeric AST were evaluated in linoleic acid(LA), liver mitochondria(LM) and red blood cell oxidation model. The chiral structure of AST from Haematococcus pluvialis and Phaffia rhodozyma was almost S and R stereoisomers. Synthetic AST is a statistical mixture(M, S ∶ meso ∶ R = 1 ∶ 2 ∶ 1).Different stereoisomeric AST showed remarkably inhibition of lipid peroxidation, including auto-oxidation of LA, the LA oxidation product malondialdehyde(MDA) content induced by Fe2+, the LM oxidation product MDA content induced by AAPH, the erythrocyte hemolysis induced by H2 O2.The anti-lipid peroxidation activity of S and R AST was significantly higher than that of M AST. The MDA inhibition rate and swelling inhibition rate of LM treated by S AST were significantly higher than those of R AST. The activities of antioxidant enzymes(SOD and GSH-PX) showed no significant differences among different stereoisomers, indicating that anti-oxidase enzyme system was not the cause of differences in anti-lipid peroxidation activity among different stereoisomers. Our results provided mechanistic information on the anti-lipid peroxidation activity of different stereoisomeric AST, which is useful for rational utilization of AST in functional food.
