摘 要:采用高效凝胶渗透色谱(HPGPC)测定β-葡聚糖含量,对其测定条件、准确度、重复性和回收率等进行研究,并采用该方法对麦类β-葡聚糖含量进行测定,研究其相对分子质量分布规律。结果表明:该方法具有准确性高、重复性好、简单易行等优点,可用于麦类中β-葡聚糖含量的测定。采用该方法对麦类中β-葡聚糖含量测定结果显示,不同品种小麦、燕麦、荞麦β-葡聚糖含量分别为0.42%~0.61%,2.16%~3.43%,1.17%~1.79%。该研究还表明,麦类中β-葡聚糖的含量及相对分子质量的分布随品种、产地的不同而有所差异。High performance gel permeation chromatography(HPGPC)was used to determine the content ofβ-glucan.The determination condition,accuracy,repeatability and recovery rate of the method were investigated,and the method was used to determine the content ofβ-glucan in different grains to study the distribution regularities of relative molecular weight.The results show that the method is accurate,simple and has a good reproducibility,so it can be used in the determination ofβ-glucan in different grains.The content ofβ-glucan in different grains was determined by this method.In different varieties of wheat,oat and buckwheat,the content ofβ-glucan is 0.42% ~0.61%,2.16% ~3.43%,1.17% ~1.79%,respectively.In addition,this study shows that the content and relative molecular weight distribution ofβ-glucan in wheat,oat and buckwheat from different growing areas and varieties are discrepant.
