摘  要:【目的】分离海洋来源的琼胶酶产生菌,确定其分类地位,研究其所产琼胶酶粗酶的酶学性质及酶解产物的抗氧化活性,为琼胶酶的进一步开发应用提供理论依据。【方法】利用以琼胶为唯一碳源的筛选培养基筛选分离产琼胶酶的菌株;采用16S r RNA基因序列分析确定菌株的分类地位;研究菌株所产琼胶酶粗酶的酶学性质;通过测定酶解产物的还原能力和清除自由基ABTS·+的能力,分析酶解产物的抗氧化活性。【结果】分离到一株产胞外琼胶酶的菌株JMUAZ5,根据16S r RNA基因序列分析,该菌株归属于弧菌属(Vibrio sp.);菌株JMUAZ5琼胶酶粗酶具有良好的底物专一性;最适反应温度为40°C,在20°C和25°C条件下稳定;最适反应p H值为8.0,在p H 6.0~10.0范围内稳定;Ca^(2+)和Al^(3+)对酶活性具有强烈促进作用,而Zn^(2+)和Fe^(3+)对酶具有强烈抑制作用;琼胶酶粗酶对测试的抑制剂、去垢剂和变性剂均具有好的抗性;酶解产物琼胶低聚糖具有还原能力和清除ABTS·+的能力。【结论】海洋细菌弧菌属JMUAZ5可产胞外琼胶酶,酶解产物有一定的抗氧化活性,具有潜在应用价值。[Objective] The aim is to identify a marine agarase-producing bacterium, characterize its crude enzyme and analyse the antioxidant activity of the enzymatic hydrolysate. [Method] The agar-degrading bacteria were isolated from marine mud by screening medium with agar as the sole carbon source. The strain was identified by the analyses of 16 S r RNA gene sequence. Its crude enzyme was characterized and the antioxidant activity of the enzymatic hydrolysate were analysed by detecting the reducing ability and scavenging power on ABTS radicals. [Result] 16 S rRNA phylogenetic analyses showed that the agarase-producing strain JMUAZ5 belonged to the genus Vibrio sp., which could produce extracellular agarase, its crude enzyme had significant substrate specificity. The optimum temperature and pH of strain JMUAZ5 agarase were 40 ℃ and 8.0, respectively. The enzymatic activity was stable at 20 ℃ and 25 ℃. It also showed stability across a pH range of 6.0-10.0. Ca^2+and Al^3+could strongly promote the agarase activity. Whereas, Zn^2+ and Fe^3+ significantly inhibited the enzyme activity. The crude enzyme had good resistance to detect inhibitors, detergents and denaturant. In addition, the enzymatic hydrolysate, agaro-oligosaccharides, had reducing ability and scavenging power on ABTS radicals. [Conclusion] Marine bacterium Vibrio sp. JMUAZ5 could produce extracellular agarase, its enzymatic hydrolysate had antioxidant activity and potential application values.
